This rabbit polyclonal antibody targets Checkpoint Kinase 2 (Chk2), a serine/threonine protein kinase that is a key effector of the DNA damage response pathway. Chk2 is phosphorylated by ATM on threonine 68 in response to double-strand DNA breaks and in turn phosphorylates substrates including p53, Cdc25A, Cdc25C, and BRCA1, driving cell-cycle arrest, homology-directed repair, or apoptosis when genome integrity cannot be restored.

From Antibodies.com, the reagent is supplied as a 100 µl volume at 1 mg/ml in PBS containing 0.1% sodium azide and 50% glycerol as cryoprotectant. The unconjugated IgG is delivered in a vial format suitable for aliquoting on arrival to limit freeze-thaw cycles, with working aliquots stored at -20 °C for long-term stability and a daily-use stock kept refrigerated at 4 °C.

Validated applications are Western blot (WB), immunohistochemistry (IHC), immunocytochemistry (ICC), immunofluorescence (IF), and immunoprecipitation (IP). Reactivity has been confirmed against Chk2 from human, mouse, and rat, so the antibody can be carried across cell-line panels, xenograft tissue sections, and primary rodent samples within a single project. Because the reagent is polyclonal, the paratope pool recognizes several epitopes on the Chk2 protein, which is useful when monitoring phosphorylation-induced band shifts on Western blot or when pulling down endogenous Chk2 complexes from cell lysate for downstream mass spectrometry or co-immunoprecipitation studies.

Researchers studying ATM/ATR signaling, radiation biology, BRCA1-associated genome stability, and the response of cancer cells to DNA-damaging chemotherapy commonly use this reagent to track pathway activation through phospho-Chk2 readouts or to localize activated Chk2 in fixed tissue and cell preparations. The unconjugated format leaves fluorophore, biotin, or HRP pairing entirely in the hands of the end user, supporting both chromogenic immunohistochemistry and multiplexed fluorescence workflows.
