Activated by ATM through phosphorylation at serine 33 and serine 35 in response to DNA double-strand breaks, CHEK2 (Checkpoint Kinase 2) is a central effector of the DNA damage response, and this rabbit polyclonal antibody from Antibodies.com selectively detects the dual phospho-epitope that marks the activated kinase. The reagent is supplied as a 100 µl liquid volume of unconjugated IgG raised in rabbit, is validated for Western blot on human samples, and is compatible with HRP-, fluorophore-, or biotin-labeled secondary reagents of the user's choice.

Once modified at S33 and S35, CHEK2 propagates the damage signal to downstream substrates such as p53, BRCA1, and the CDC25 phosphatase family, enforcing cell cycle arrest at the G1/S and G2/M checkpoints. Researchers therefore use this antibody to monitor checkpoint activation after genotoxic insults such as ionizing radiation, topoisomerase inhibitors, or PARP inhibition, and to study CHEK2 signaling in the context of tumor suppression, genomic stability, and cellular chemosensitivity in human cells.

The polyclonal host response provides robust affinity for the dual phospho-epitope on the activation loop, allowing clear detection of activated CHEK2 in cultured human cell lysates and tissue-derived samples by Western blot. For best results, aliquot upon arrival to limit freeze-thaw cycles and store the working stock refrigerated at 4 degrees C for short-term use.
