Phosphorylation of cyclin-dependent kinase 1 (Cdk1/Cdc2) at threonine 14 is one of two inhibitory N-terminal marks that restrain cells at the G2/M boundary until DNA replication is finished and any checkpoint signal is cleared. The Antibodies.com polyclonal antibody supplied here recognizes this phosphorylated residue in the context of full-length Cdk1, so phosphorylation dynamics can be read directly against total kinase levels on matched lanes or sections.
This unconjugated reagent is provided as 100 µl at 1 mg/ml in PBS with 0.1% sodium azide and 50% glycerol, and is validated for Western blot, immunohistochemistry, and immunofluorescence on Human, Mouse, and Rat samples. Because the host species is rabbit and the reagent is polyclonal, it tolerates various fixation and permeabilization protocols used in IHC and IF, and pairs cleanly with anti-rabbit secondary antibodies carrying HRP, fluorophore, or biotin labels.
Typical use cases include monitoring G2 arrest after DNA damage, tracking release from a checkpoint block into mitosis, and comparing phospho-T14 signal across synchronized cell populations such as double-thymidine or nocodazole-arrested cultures versus asynchronous controls. The threonine-14 mark is laid down by Wee1 and Myt1 kinases and removed by Cdc25 phosphatases, so this reagent fits studies of upstream checkpoint signaling, ATM/ATR pathway activity, and the kinetics of mitotic entry in oocytes, somatic cells, and tissue sections.
Store the vial at -20 °C in working aliquots and limit freeze-thaw cycles; the glycerol in the formulation protects activity through routine handling. Allow the reagent to reach room temperature before opening to limit condensation, then dilute into standard antibody diluent for the application of choice.
