This rabbit polyclonal reagent from Antibodies.com targets Cadherin-23 (CDH23), a calcium-dependent adhesion protein that forms the transient lateral links and tip links of inner-ear hair-cell stereocilia and is also detected in retinal photoreceptors. The antibody is supplied as 100 µL of a 1 mg/mL stock in PBS with 0.1% sodium azide and 50% glycerol, unconjugated so it can be matched to a secondary detection reagent of choice for chromogenic or fluorescence workflows. It has been validated for western blot and immunohistochemistry on human, mouse, and rat samples, making it relevant to research on Usher syndrome type 1D, DFNB12 non-syndromic hearing loss, and hair-cell bundle mechanotransduction. The polyclonal format increases the likelihood of recognising more than one CDH23 epitope, which is helpful on tissue sections where the large extracellular domains may be partially cleaved. Storage at -20 °C in working aliquots is recommended to limit freeze-thaw cycles of the glycerol stock, and each vial is intended for single-use preparation in standard blocking buffer for immunoblotting or IHC protocols. Researchers studying otic development, stereocilia turnover, or retinal cadherin function typically use this reagent at 1-5 µg/mL for western blot and at 5-10 µg/mL for immunohistochemistry on paraffin- or cryo-embedded sections.
