Targeting the CD4 glycoprotein expressed on helper T lymphocytes, this unconjugated Mouse monoclonal antibody (clone 10H8) is supplied by Antibodies.com as 100 µl of stock at 1 mg/ml in phosphate buffered saline (PBS) with 0.05% sodium azide at approximately pH 7.3. The reagent has been validated for Western blot applications and cross-reacts with Human, Mouse, and Rat samples, enabling side-by-side comparison of CD4 expression across these species in primary cell lysates, tissue homogenates, and immunoblotting workflows.

CD4 functions as a co-receptor for MHC class II-restricted antigen recognition and is required during T-cell receptor engagement at the immune synapse. In addition to helper T cells, CD4 is detected at lower levels on monocytes, macrophages, and some dendritic cell subsets, so the 10H8 clone is useful for characterizing CD4+ populations and for monitoring CD4 downregulation in relevant viral infection models. Sodium azide in the storage buffer inhibits microbial growth and should be removed by buffer exchange or dialysis before any live-cell or in vivo application.

For Western blot, dilute the stock in TBST containing 5% non-fat milk or BSA according to standard immunoblotting protocols, incubate with transferred membranes for 1-2 hours at room temperature or overnight at 4 °C, and detect bound antibody with an HRP-conjugated anti-mouse secondary. Short-term storage refrigerated preserves activity; for extended storage, aliquot the antibody to limit freeze-thaw cycles and protect from light. Researchers working with peripheral blood mononuclear cells, splenocytes, thymus, or lymphoid tissue routinely deploy this clone to track CD4+ populations and to confirm immune phenotypes in transgenic and knockout mouse models.
