The Antibodies.com polyclonal reagent targets human CD14, a glycosylphosphatidylinositol-anchored surface protein best known as the LPS co-receptor that delivers bacterial lipopolysaccharide to the TLR4/MD-2 complex on monocytes and macrophages. Raised in rabbit and purified as total IgG, this unconjugated antibody is supplied at 50 µl and is validated for Western Blot detection of endogenous CD14 in human cell and tissue lysates.

CD14 exists in two biologically relevant forms: the GPI-anchored membrane form (mCD14) on myeloid cells and a soluble form (sCD14) shed into plasma during inflammation, which both contribute to innate immune signaling. Polyclonal antibodies such as this one bind multiple epitopes on the target, which can be useful when probing denatured protein on a membrane but should be considered if cross-reactivity with closely related proteins is a concern. Because the host species is rabbit and the isotype is IgG, secondary anti-rabbit IgG reagents conjugated to HRP, fluorophores, or biotin can be applied for standard WB chemiluminescent or fluorescent detection workflows.

Typical use begins with sample preparation in standard RIPA or similar lysis buffer, separation by SDS-PAGE, transfer to nitrocellulose or PVDF, blocking, and overnight primary incubation. This reagent has not been validated by Antibodies.com for flow cytometry, immunohistochemistry, or ELISA in the current datasheet, so it is best suited to researchers specifically wanting a WB-only reagent or to those willing to validate additional applications themselves. For research use only.
