Cyclin B1 turnover during mitotic exit and after DNA damage is gated by phosphorylation at serine 147, which generates a phosphodegron recognized by the SCFβ-TrCP ubiquitin ligase. This unconjugated rabbit polyclonal IgG reagent from Antibodies.com was raised against human Cyclin B1 to selectively detect the phospho-S147 form, allowing researchers to monitor modification of the cyclin independently of changes in total protein abundance.

Supplied at 200 µl, the antibody is validated for western blot, immunohistochemistry, and immunofluorescence on human samples. The polyclonal format offers multiple contacts against the modified epitope, and the unconjugated format leaves choice of HRP, fluorophore, or biotin secondary labels to the user.

Research uses include tracking mitotic entry and the G2/M transition in synchronized cell populations, mapping CK1-driven phosphorylation of the S147 degron, comparing the phospho-S147 signal in untreated versus genotoxin-treated cells, and assessing aberrant Cyclin B1 turnover in tumor-derived cell lines and tumor tissue where mitotic control is dysregulated.

For handling, refrigerate the reagent upon arrival, aliquot to limit freeze-thaw cycles, and protect from light when working with any downstream conjugate prepared from this antibody.
