Raised against the G650 region of β-catenin, this unconjugated polyclonal reagent is supplied by Antibodies.com as 100 µl of a 1 mg/ml stock in phosphate buffered saline with 0.05% sodium azide at approximately pH 7.2. β-catenin is a dual-function protein that anchors the cytoplasmic tail of E-cadherin to the actin cytoskeleton at adherens junctions and, when the canonical Wnt pathway is active, escapes degradation, accumulates in the cytoplasm, and translocates into the nucleus where it partners with TCF/LEF transcription factors to drive target genes such as c-Myc and cyclin D1. That central position at the intersection of cell-cell adhesion, embryonic development, stem cell maintenance, and oncogenesis is why a well-characterized detection antibody is a workhorse in pathway-focused labs.
The antibody is validated for western blot, immunohistochemistry, and immunofluorescence on human, mouse, and rat samples. In western blot it detects the expected band near 92 kDa, while in fixed sections and cultured cells it produces cytoplasmic, membranous, and nuclear staining depending on Wnt pathway status, which is useful for comparing tumor versus normal tissue, mapping subcellular redistribution after pathway stimulation or inhibition, and monitoring epithelial-mesenchymal transition. Common applications include studies of colorectal and hepatocellular carcinoma, intestinal crypt homeostasis, neural crest and cardiac development, tissue regeneration, and β-catenin-driven transcriptional programs. Because reactivity is confirmed across three species, the same lot can be used for parallel work in human cell lines, mouse xenografts, and rat disease models without switching primaries.
As a polyclonal reagent, the antibody recognizes multiple epitopes on β-catenin, which improves signal in cross-linked or fixed specimens where protein conformation and phosphorylation state can mask individual epitopes. The vial should be kept refrigerated and protected from light, with small aliquots prepared to limit freeze-thaw cycles; the sodium azide preservative allows the antibody to be added directly to most staining and blotting protocols without further purification. Intended for research use only, this reagent supports investigations of Wnt signaling, cadherin-catenin adhesion complexes, and β-catenin-mediated gene regulation in cancer biology and developmental research.
