Raised in rabbit against the human Caspase-14 (CASP14) target, this unconjugated polyclonal antibody from Antibodies.com is supplied as 100 µl of ready-to-use reagent for Western blot and immunofluorescence detection. CASP14 is a non-apoptotic cysteine protease largely confined to epidermal keratinocytes, where it cleaves pro-filaggrin and contributes to the formation of the cornified envelope during terminal differentiation of the skin barrier.

Because the reagent is unconjugated, it can be paired with the researcher's preferred secondary system such as HRP, biotin, or a fluorophore, which is convenient when the same primary must serve both immunoblotting of keratinocyte lysates and imaging of human skin sections. Polyclonal reactivity against multiple epitopes on human CASP14 improves tolerance for minor sequence variation and supports detection of the full-length proenzyme as well as its processed p20 and p10 subunits that accumulate in cornified tissue.

Recommended uses include Western blot to follow CASP14 expression across a keratinocyte differentiation time course, and immunofluorescence to localize CASP14 within the granular and cornified layers of the epidermis. The 100 µl volume is sufficient for multiple pilot blots and imaging runs from a single vial; aliquot on first use and store refrigerated to preserve reagent performance across repeat experiments.
