Generated in rabbit and affinity-purified, this unconjugated polyclonal antibody from Antibodies.com targets Calpain Small Subunit 2 (CAPNS2) for Western blot applications on cell and tissue extracts of human, mouse, and rat origin. The reagent ships as 100 µl at 1 mg/ml in phosphate buffered saline (pH ~7.2) containing 15 mM sodium azide as a microbial preservative, ready for dilution directly into standard reducing Laemmli sample buffer without further preparation.

CAPNS2 is the smaller regulatory subunit of the heterodimeric calpain protease complex, partnering with catalytic large subunits (calpain-1, calpain-2, and others) to confer calcium-dependent activation and substrate selectivity. Because CAPNS2 shares homology with the more widely expressed CAPNS1 paralog, a CAPNS2-selective reagent is useful for distinguishing the two small subunits in studies of calpain-dependent cytoskeletal remodeling, myoblast fusion, neuronal plasticity, and tumor cell migration. The polyclonal format provides multi-epitope recognition of the ~28 kDa CAPNS2 band, helping reduce the impact of minor epitope variations between the species tested.

Recommended working concentration for immunoblotting is approximately 0.5-2 µg/ml, detected with an HRP-conjugated anti-rabbit secondary antibody and chemiluminescent substrate. Aliquot the vial upon arrival if repeated experiments are planned, refrigerate for short-term use, and store frozen at -20°C for long-term retention of activity. Avoid repeated freeze-thaw cycles and protect from light to preserve reagent performance.
