This rabbit polyclonal antibody from Antibodies.com is raised against the autophosphorylated alpha isoform of CaMKII, specifically recognizing the Thr286 phospho-epitope that marks the constitutively active kinase. Threonine-286 autophosphorylation is the molecular switch underlying CaMKIIα activation during calcium influx, supporting studies of synaptic plasticity, long-term potentiation, and neuronal signaling in rodent and human tissue.

The antibody is supplied at 1 mg/ml in phosphate buffered saline (PBS) pH 7.2 with 0.05% sodium azide as preservative, in a 100 µl vial. It is validated for Western blot (WB) and immunohistochemistry (IHC) applications, and shows reactivity against Human, Mouse, and Rat samples. For western blotting, researchers commonly probe membranes for the upper ~50 kDa band corresponding to CaMKIIα and confirm autophosphorylation by comparing calcium-stimulated versus basal lysates.

Aliquot upon first use to limit freeze-thaw cycles; the sodium azide-containing buffer is compatible with brief storage at 4°C for working aliquots, with the stock held at -20°C for long-term stability. The unconjugated format provides flexibility for downstream labeling, including fluorescent secondary detection in IHC and HRP-based chemiluminescence in WB. As a polyclonal reagent, it offers broader epitope coverage than monoclonal alternatives and tolerates minor conformational variation around the T286 site.
