The target is a GPI-anchored type I transmembrane isoform that catalyzes the reversible hydration of CO2 to bicarbonate and a proton, supporting pH and CO2 homeostasis in epithelial and neural tissues. Published work localises endogenous CA14 to the apical and basolateral membranes of retinal pigment epithelium, to Müller cell and astrocytic membranes in mouse retina, and to the proximal convoluted tubule of the kidney, the major segment for bicarbonate reabsorption. Validated for western blot on human and mouse samples, the reagent detects total endogenous CA14 protein and is used in studies of renal acid-base transport, retinal ion regulation, and neuronal carbonic anhydrase activity. Affinity-purified from rabbit antiserum by immunoaffinity chromatography, the polyclonal format surfaces multiple epitopes across the CA14 sequence, which is useful when probing splice variants or post-translationally modified forms of the enzyme.
