The c-Src protein is a 60 kDa non-receptor tyrosine kinase and the founding member of the Src family of kinases, a group of enzymes that couple growth factor, integrin, and G-protein-coupled receptor signaling to cytoskeletal remodeling and cell adhesion. Antibodies.com offers a rabbit polyclonal antibody raised against an epitope surrounding residue N535 of human c-Src, validated for immunohistochemistry on formalin-fixed paraffin-embedded tissue sections. The reagent is supplied at 1 mg/ml in phosphate buffered saline containing 0.05% sodium azide at approximately pH 7.2, and is packaged as a 100 µl vial.

Cross-reactivity is reported with human, mouse, and rat samples, supporting translational studies that compare Src pathway activity across species in tumor biopsies and animal disease models. Because the reagent is supplied unconjugated, it is compatible with many HRP- or fluorophore-conjugated secondary antibodies and chromogenic substrates chosen by the end user, including routine DAB-based IHC workflows. Src family kinases are frequently upregulated in epithelial and mesenchymal tumors and contribute to downstream effectors such as focal adhesion kinase, so this reagent is commonly used to map kinase distribution within the tumor microenvironment, to investigate Src-dependent signaling in wound healing and bone remodeling studies, and to probe Src activity in neural tissue where the kinase regulates NMDA receptor function and axon guidance.

Recommended applications include antigen retrieval on FFPE sections followed by overnight primary incubation; endogenous peroxidase quenching is appropriate because the buffer contains no proprietary blocking proteins. Sodium azide is included as a preservative, so the antibody is compatible with standard 4 °C short-term handling but should be diluted in azide-free buffer if it will be used in live-cell or HRP-conjugate workflows downstream.
