The c-Met receptor tyrosine kinase, also called MET or HGFR, is activated by hepatocyte growth factor (HGF) and signals through tyrosine residues in both the kinase activation loop (Y1234 and Y1235) and the C-terminal multifunctional docking site, which contains Y1349 and Y1356. Among these sites, phosphorylation at Y1349 is the critical event that creates the binding platform for downstream adaptors such as Gab1, Grb2, and STAT3, propagating signals that control proliferation, scattering, migration, and invasive growth. Because Y1349 phosphorylation correlates with active receptor signaling, antibodies that recognize only this phospho-form are widely used as readouts of pathway activation in cancer research, where aberrant MET signaling is implicated in hepatocellular carcinoma, gastric cancer, non-small cell lung cancer, and other tumor types.

This reagent is a rabbit polyclonal antibody from Antibodies.com raised against the phospho-Y1349 epitope of human c-Met. It is supplied as an unconjugated polyclonal preparation at 1 mg/ml in phosphate buffered saline (PBS) with 0.05% sodium azide at approximately pH 7.2, in a 100 µl vial. The antibody detects the phosphorylated receptor in samples of human, mouse, and rat origin, supporting cross-species comparison of MET pathway activation in cell line models and in tissue specimens. The validated application is Western blot, in which the antibody is applied to whole-cell lysates prepared with standard phosphatase inhibitor cocktails (such as cocktails containing sodium orthovanadate and sodium fluoride) so that the Y1349 phospho-epitope is preserved during sample handling.

For storage, keep the vial refrigerated and protect from light; aliquot the antibody if extended storage is needed to limit freeze-thaw cycles, and store prepared lysates frozen until blotting. Because the antibody is unconjugated, it can be paired with a secondary reagent of the researcher's choice for chemiluminescent or fluorescent detection on Western blot membranes.
