Raised in rabbit against the BCL2 antagonist/killer 1 (BAK1) protein, this polyclonal antibody from Antibodies.com targets a pro-apoptotic BCL-2 family member that oligomerizes within the outer mitochondrial membrane to drive cytochrome c release during the intrinsic apoptosis pathway. The unconjugated reagent is supplied as 100 µl aliquots and is validated for western blot, immunohistochemistry, and immunofluorescence workflows on human, mouse, and rat specimens.

BAK1 functions as a critical executioner of programmed cell death. In response to cellular stress, it undergoes conformational activation at the mitochondrial outer membrane, where it forms homooligomers that permeabilize the membrane and trigger caspase activation. Loss or mutation of BAK1 has been associated with altered apoptotic responses in cancer cell lines, making this reagent relevant to studies of tumor cell survival, chemotherapeutic sensitivity, and mitochondrial pathway regulation.

For typical use, apply the antibody to PVDF or nitrocellulose membranes for immunoblotting of BAK1 expression (an approximately 25 kDa band), to fixed tissue sections for IHC localization, or to cultured cells for IF visualization of mitochondrial staining patterns. The 100 µl vial supports multiple experiments; aliquot on receipt and keep refrigerated to limit freeze-thaw cycles of the polyclonal stock.
