This rabbit polyclonal antibody from Antibodies.com specifically recognizes Bad (BCL2-associated agonist of cell death) when phosphorylated at serine 155. Phosphorylation at S155 is a key regulatory event in apoptosis control: kinases such as Akt/PKB phosphorylate Bad at this site, driving sequestration of the protein by 14-3-3 scaffolds in the cytoplasm and thereby preventing Bad from antagonizing anti-apoptotic Bcl-2 family members at the mitochondrial outer membrane. The reagent is supplied as 50 µl of unconjugated IgG, validated for Western blot and immunohistochemistry workflows, and reactive against human, mouse, and rat samples. Researchers use this antibody to track survival signaling downstream of PI3K-Akt, to map phosphorylation dynamics in cancer cell lines, neuronal injury models, and cytokine-treated primary cultures, and to distinguish the inactive, cytoplasmic pool of Bad from the mitochondrial, pro-apoptotic form. Because the reagent is phospho-specific, it reports kinase activity rather than total Bad abundance, making it a useful tool for studies of growth factor signaling, oxidative and genotoxic stress responses, and small-molecule modulators of the apoptotic machinery.
