This unconjugated rabbit polyclonal IgG reagent from Antibodies.com is raised against the catalytic B1 subunit of the vacuolar-type H+-ATPase (V-ATPase), encoded by ATP6V1B1. The target drives ATP-driven proton translocation across membranes and is enriched in renal alpha-intercalated cells of the cortical collecting duct, in the stria vascularis and endolymphatic epithelium of the inner ear, and in epididymal clear cells, which explains the well-characterized link between biallelic ATP6V1B1 loss-of-function variants and autosomal recessive distal renal tubular acidosis with progressive sensorineural hearing loss (dRTA2, OMIM 192132).

Supplied as 100 µl of liquid IgG in a single-use vial, the antibody is validated for Western blotting, immunohistochemistry, and immunofluorescence on human tissue sections and cell preparations. On Western blots of human kidney or HeLa lysates, signal appears near 56 kDa, consistent with the calculated mass of the mature B1 subunit. In immunohistochemistry, staining localizes to the apical cytoplasm of intercalated cells, while in immunofluorescence preparations an apical-membrane and punctate cytoplasmic distribution is observed in polarized epithelia.

Routine handling calls for brief equilibration to room temperature before use, aliquoting to avoid repeated freeze/thaw cycles, and storage at -20°C in the supplied phosphate-buffered saline formulation with 50% glycerol and 0.02% sodium azide. The reagent is for research use only and is not intended for diagnostic or therapeutic applications.
