ATP5B is the catalytic beta subunit of the F1 complex of mitochondrial ATP synthase, the rotary enzyme that couples the proton gradient across the inner mitochondrial membrane to ATP synthesis via oxidative phosphorylation. This polyclonal reagent from Antibodies.com was raised in rabbit and recognises an epitope on the human, mouse, and rat proteins, supporting comparative work across standard mammalian systems. The antibody is supplied as 100 µl of a 1 mg/ml stock in PBS with 0.1% sodium azide and 50% glycerol, so it can be held at 4 °C for routine use and stored at -20 °C in working aliquots to limit freeze-thaw damage to the stock. It is validated for Western blot on whole-cell and mitochondrial lysates, where the dominant band near 56 kDa makes it a common loading control for normalising mitochondrial protein content, and for immunohistochemistry on fixed tissue sections to map expression of the oxidative phosphorylation machinery. Because the protein is nuclear-encoded and imported into mitochondria, the reagent is also suited to subcellular localisation studies, mitochondrial biogenesis work, and disease models of energy metabolism dysfunction.
