A central cysteine protease in macroautophagy, ATG4B cleaves pro-LC3 and GABARAP family proteins at their C-terminus, exposing the C-terminal glycine required for conjugation to phosphatidylethanolamine and subsequent recruitment onto autophagosomal membranes. This Antibodies.com reagent is a rabbit polyclonal antibody raised against the human protein, validated for western blot detection of endogenous ATG4B in cell and tissue lysates from human, rat, and mouse origins.

Each vial ships with 100 µl of unconjugated antibody at approximately 1 mg/ml, formulated in phosphate buffered saline at pH 7.3 with 0.05% sodium azide as preservative. Because the host is rabbit and the target sequence is conserved across mammals, the reagent is suited to parallel immunoblotting workflows comparing autophagic activity across species, and it is commonly paired with anti-LC3 and anti-p62 antibodies to monitor autophagosome lipidation and substrate turnover. The unconjugated format leaves reporter choice to the researcher.

Refrigerate the vial for short-term storage; for longer-term holding, aliquot to limit freeze-thaw cycles and freeze. A polyclonal mixture targeting multiple ATG4B epitopes is particularly useful for tracking expression of the protease itself in studies of mitophagy, ER-phagy, and selective autophagy research use, complementing downstream readouts of autophagic flux under basal, induced, or inhibited conditions.
