ATF6 is a 90 kDa transmembrane transcription factor anchored in the endoplasmic reticulum that acts as a sensor of misfolded protein accumulation and drives the unfolded protein response. Under ER stress, ATF6 translocates to the Golgi, where site-1 and site-2 proteases cleave it, releasing an approximately 50 kDa amino-terminal fragment that enters the nucleus and induces transcription of ER chaperones and folding enzymes such as BiP/GRP78 and XBP1.
