Raised in rabbit against a synthetic phosphopeptide spanning the threonine 71/threonine 53 region of human ATF2, this affinity-purified polyclonal IgG from Antibodies.com selectively recognizes the activated, dual-phosphorylated form of activating transcription factor 2 and does not detect the non-phosphorylated protein. The reagent is supplied as an unconjugated liquid in a 200 µl volume and is validated for western blot and immunohistochemistry on human, mouse, and rat samples, with a target band near 52 kDa on immunoblots of stressed-cell lysates.

Phosphorylation at Thr71 and Thr53 is a direct readout of stress-activated kinase activity, since these two residues are modified by JNK and p38 MAPK in response to genotoxic damage, oxidative stress, inflammatory cytokines, and UV irradiation. Researchers therefore use this antibody to monitor time-course activation of the SAPK/JNK and p38 pathways following stimuli such as anisomycin, sorbitol, cisplatin, or tumor necrosis factor, and to compare basal versus induced phospho-ATF2 levels between wild-type and kinase-deficient or knock-out backgrounds. The antibody is also applied to formalin-fixed paraffin-embedded sections to chart ATF2 activation status within tumor tissue microenvironments, including breast carcinoma and melanoma specimens, and to correlate nuclear phospho-ATF2 staining with downstream AP-1/CRE-driven gene expression programs in disease models.

Among ATF/CREB-family antibodies, the reagent is notable for its narrow specificity: characterization data show that it does not cross-react with phosphorylated c-Jun, CREB, or other related transcription factors, which makes it suitable for cleanly resolving the ATF2 contribution to AP-1 complexes in chromatin immunoprecipitation and reporter assays. For storage, the product is shipped cold and should be aliquoted and stored at -20°C to limit freeze-thaw cycles. The reagent is intended for research use only and is not approved for diagnostic applications.
