A phosphorylation-state-specific rabbit polyclonal IgG from Antibodies.com is raised against a synthetic peptide derived from human ATF2 (amino acids 79-128) and selectively recognizes the dually phosphorylated form of the transcription factor at Ser112 and Ser94, the residues modified by stress-activated JNK and p38 MAPK kinases. In western blot, the reagent yields a band near the 52 kDa molecular weight marker; in immunohistochemistry, it produces clean nuclear and cytoplasmic staining on Human, Mouse, and Rat tissue sections. Each vial contains 200 µl of liquid antibody purified from rabbit serum by antigen affinity chromatography, supplied in phosphate buffered saline (pH 7.4) with 150mM NaCl, 0.02% sodium azide, and 50% glycerol as a cryoprotectant. Recommended starting dilutions are 1:500 to 1:1000 for western blot and 1:50 to 1:100 for IHC. The product is shipped at 4°C; upon arrival it should be aliquoted and stored at -20°C, with repeated freeze-thaw cycles avoided to preserve recognition of the phospho-epitopes. Researchers mapping UV- or genotoxin-triggered kinase cascades, cytokine-driven transcriptional programs, and chromatin-associated regulatory mechanisms can use this reagent to confirm endogenous phosphorylation events in cell lines and primary samples.
