Description

A rabbit polyclonal antibody from Antibodies.com, unconjugated and supplied at 1 mg/ml in PBS with 0.1% Sodium Azide and 50% glycerol. The reagent is raised against Artemis phosphorylated at Serine 516 and has been validated for Western blot (WB) and immunohistochemistry (IHC) on human, mouse and rat samples.

Specificity is directed at endogenous levels of the Artemis protein only when phosphorylated at Ser516, as indicated by the synthetic phosphopeptide immunogen derived from human Artemis around the S516 site. The antibody was affinity-purified from rabbit antiserum by affinity-chromatography using epitope-specific immunogen, with purity greater than 95% by SDS-PAGE; the apparent molecular weight is approximately 78 kDa.

Artemis contributes to the preservation of double-strand DNA breaks by processing damaged DNA ends. It functions as a single-strand-specific 5' to 3' exonuclease that forms a complex with the 469 kDa DNA-dependent protein kinase (DNA-PKcs). Upon phosphorylation by DNA-PKcs, Artemis acquires endonucleolytic activity on 5' and 3' overhangs and hairpins, activities essential for V(D)J recombination and nonhomologous DNA end joining.
