Aquaporin-4 (AQP4) is a water-selective channel protein concentrated in astrocyte end-feet at the blood-brain barrier, where it regulates cerebrospinal fluid homeostasis and is the principal autoantigen targeted in neuromyelitis optica spectrum disorder (NMOSD). This unconjugated rabbit polyclonal primary antibody from Antibodies.com recognizes the human, mouse, and rat orthologs and is validated for western blot detection of AQP4 in brain, spinal cord, kidney, and transfected cell lysates.

The reagent is supplied as a 50 µl vial at 1 mg/ml in phosphate buffered saline with 0.05% sodium azide at approximately pH 7.3. On denaturing western blots of whole-tissue lysate, the antibody detects the major AQP4 isoforms, allowing researchers to map isoform distribution between tissues. Investigators working on glial water transport, blood-brain barrier integrity, NMO-IgG serology, astrocyte biology, and animal models of cerebral edema and ischemic stroke use this reagent to confirm AQP4 expression across different sample types.

For handling, store refrigerated, aliquot upon first use to limit freeze-thaw cycles, and pair with an HRP-conjugated anti-rabbit secondary. A typical working concentration on western blot is 1-2 µg/ml in 5% non-fat milk in TBST, with chemiluminescent detection of the labeled band.
