Raised in rabbit as a polyclonal IgG preparation, this unconjugated primary antibody from Antibodies.com binds the amyloid precursor protein (APP), a type I transmembrane glycoprotein whose proteolytic processing by α-, β-, and γ-secretases generates the amyloid-beta peptides linked to Alzheimer's disease pathology. The reagent is validated for western blot (WB) detection of human and mouse APP and, in immunoblotting workflows, can resolve full-length holo-APP migrating at approximately 95–100 kDa together with its soluble APP, C-terminal fragments, and amyloid-beta cleavage products following SDS-PAGE separation of cell or tissue lysates. Supplied as 100 µl of liquid, the antibody offers flexibility in downstream detection chemistry: because it is unconjugated, researchers can pair it with HRP-conjugated anti-rabbit secondaries for chemiluminescent readouts or with fluorophore-conjugated secondaries for infrared or fluorescent imaging systems. The polyclonal format increases the likelihood of recognizing multiple APP isoforms, including APP695, which predominates in neurons, and APP751/APP770, which are more broadly expressed across peripheral tissues, so a single blot can survey the major splice variants in parallel. Suggested lysate inputs of roughly 20–50 µg total protein separated on 8–10% polyacrylamide gels and probed at working dilutions in the 1:500 to 1:1000 range support clean detection of endogenous APP levels. Typical applications include tracking amyloidogenic versus non-amyloidogenic cleavage, monitoring APP trafficking and processing in neuronal cell culture, and comparing APP expression between wild-type and transgenic mouse models of neurodegeneration. Store the antibody refrigerated at 2 to 8 degrees C in working aliquots to minimize repeated freeze-thaw cycles, and short-term handling on ice is recommended immediately before immunoblotting.
