The target antigen APP (Amyloid Precursor Protein) is a type I transmembrane glycoprotein encoded on human chromosome 21. Sequential cleavage by β- and γ-secretases liberates the amyloid-β peptide that accumulates in Alzheimer's disease plaques, whereas α-secretase cleavage within the Aβ sequence releases a soluble, non-amyloidogenic fragment. Alternative splicing of the APP transcript yields several isoforms; APP695 predominates in neurons, while APP751 and APP770 are expressed more broadly across peripheral tissues.

This Antibodies.com polyclonal reagent is raised in rabbit, isotype IgG, and is supplied unconjugated in a 50 µl vial. It is validated for Western blot and immunohistochemistry on human samples, making it a practical tool for probing APP expression in cell lysates and in fixed brain sections. In neuroscience research, antibodies of this class are used to map APP distribution in normal neurons, to monitor expression changes in Down syndrome models (where the trisomy 21 locus drives elevated APP levels), and to characterize processing of familial Alzheimer's disease mutations in patient-derived samples.
