Anti-AKT1 (phospho T450) Antibody

Antibodies.com supplies a rabbit polyclonal IgG antibody specific for AKT1 phosphorylated at Thr450. The unconjugated reagent is supplied as a liquid in Phosphate Buffered Saline (pH 7.3) containing 50% glycerol and 0.02% sodium azide, and has been affinity purified. It is reactive with human samples and validated for Western blot at a recommended dilution range of 1:500–1:1,000.

The immunogen used to raise the antibody is a synthetic phosphorylated peptide centred on T450 of human Akt1 (accession NP_005154.2), corresponding to the sequence TITPP. The target protein has a reported molecular weight of 60 kDa. The product is shipped at 4°C and should be aliquoted upon arrival and stored at -20°C, with freeze/thaw cycles avoided.

A representative Western blot demonstrates detection of phospho-AKT1 (Thr450) in 293 cells stimulated with 100 nM insulin for 10 minutes and in HeLa cells stimulated with 100 ng/ml EGF for 30 minutes, both following overnight serum-starvation. The antibody was used at 1:1,000 dilution with 25 µg of lysate per lane, and signal was developed with an ECL Basic Kit.
