Affinity-purified reagent from Antibodies.com provides isoform-resolved detection of the Akt1 and Akt3 serine/threonine kinases in a single primary antibody. Raised in rabbit and supplied unconjugated in a 100 µl vial, the polyclonal IgG recognizes conserved epitopes within AKT1 (PKBα) and AKT3 (PKBγ), letting researchers follow either or both isoforms from the same blot.

Verified applications include Western blot, immunohistochemistry, and ELISA. On Western blot the antibody resolves the characteristic ~56 kDa band corresponding to Akt1 and the equivalent Akt3 signal, with reactivity confirmed against human, mouse, and rat samples. Immunohistochemistry on formalin-fixed tissue produces cytoplasmic staining consistent with PI3K pathway activation in tumor, brain, and metabolic tissues, and ELISA-based assays support quantitative work where a labeled secondary is preferred.

Each lot is affinity purified and dialyzed into a glycerol-containing storage buffer. Aliquot on receipt and store at -20°C, limiting freeze-thaw cycles and protecting from light to preserve activity. The unconjugated format offers flexibility to conjugate the antibody to HRP, biotin, Alexa Fluor dyes, or gold nanoparticles as needed, and to titrate it for immunoprecipitation, immunofluorescence, or chromatin workflows without prior buffer exchange.

Akt1 and Akt3 are key downstream effectors of PI3K that govern cell survival, proliferation, glucose metabolism, and protein synthesis through mTORC1 and GSK3β phosphorylation. Separating AKT1-driven tumor proliferation from the brain-enriched roles of AKT3 in neuronal development, insulin signaling, and cardiac hypertrophy usually requires isoform-resolved detection, which is why a dual-specific polyclonal is often used alongside — or as a substitute for — separate monoclonal primaries.
