AKR1D1 is the human gene encoding the 37 kDa NADPH-dependent steroid 5β-reductase that catalyzes the stereospecific reduction of Δ⁴-3-ketosteroids during primary bile acid biosynthesis and hepatic steroid clearance. The Antibodies.com polyclonal antibody is raised in rabbit as an unconjugated IgG reagent and detects endogenous AKR1D1 protein in human, mouse, and rat samples, supporting cross-species studies of liver metabolism, congenital bile acid synthesis defect type 2 (CBAS2), and steroid hormone catabolism.

Validated for western blot, the reagent performs well on mouse liver lysates and human hepatocyte-derived cell lines, where the target is most strongly expressed. The enzyme's calculated mass of 37 kDa and observed migration near 33–37 kDa on SDS-PAGE make it straightforward to identify by molecular weight. Substrates include intermediates such as 7α-hydroxycholest-4-en-3-one and 17β-hydroxyestr-4-en-3-one, and the enzyme's activity is sensitive to feedback inhibition by chenodeoxycholic acid and ursodeoxycholic acid.

Each vial is supplied as a 50 µl liquid aliquot in PBS with 50% glycerol and 0.05% sodium azide at pH 7.3. Shipped refrigerated for stability, the antibody should be aliquoted upon arrival and stored at -20 °C, with protection from light and repeated freeze-thaw cycles avoided to preserve binding activity. For research use only, the polyclonal format recognizes multiple epitopes on AKR1D1, strengthening signal in tissues where target abundance is limited.
