Raised in rabbit against the human Autoimmune Regulator (AIRE) transcription factor, this polyclonal reagent from Antibodies.com is supplied as 100 µl of unconjugated whole antiserum for western blot and immunofluorescence workflows on human and mouse samples. AIRE drives the ectopic expression of tissue-restricted antigens in medullary thymic epithelial cells, a process central to clonal deletion of self-reactive thymocytes and the establishment of central immunological tolerance. Loss-of-function variants in AIRE underlie APECED (also called APS-1), a monogenic syndrome defined by multi-organ autoimmunity, chronic mucocutaneous candidiasis, and ectodermal dystrophy, which is why antibodies against this target are used in studies of thymic selection, peripheral tolerance checkpoints, and disease-associated autoantibody panels.

The reagent is validated for the applications listed in the product sheet: WB for protein-level detection in cell and tissue lysates, and IF for subcellular localization in cultured cells or thymic sections. Researchers commonly apply it to mTEC cultures, thymic cryosections, and AIRE-transfected overexpression systems, with reactivity confirmed against both human and mouse orthologues. The unconjugated format allows flexible pairing with custom fluorophore or enzyme labels, and the 100 µl vial provides enough material for multiple blotting or staining runs. Aliquot on receipt, store frozen to limit freeze-thaw cycles, and protect from light if a downstream fluorescent conjugate is later attached.
