Raised in rabbit against the 47 kDa actin-related protein 3 subunit of the Arp2/3 complex, this unconjugated polyclonal reagent from Antibodies.com is supplied at 100 µl per vial and validated for Western blot and immunohistochemistry on human, mouse, and rat samples. ARP3 is one of seven proteins that make up the Arp2/3 complex, where it works together with Arp2 and a nucleation-promoting factor to seed branched actin filament networks at the leading edge of motile cells and at sites of pathogen-induced actin reorganization.

For Western blot, the antibody recognizes endogenous ARP3 near its predicted molecular weight of roughly 50 kDa, producing a single predominant band in whole-cell lysates of common human and rodent lines. In immunohistochemistry, it yields cytoplasmic and cortical staining consistent with the protein's known distribution along the actin cytoskeleton, near focal adhesions, and within lamellipodia and invadopodia of adherent cultures. Cross-reactivity with the mouse and rat orthologs allows parallel comparison between engineered mammalian cell lines and rodent tissue sections.

Aliquot on first thaw and store at -20°C in working volumes to limit freeze-thaw cycles, since polyclonal sera lose titer faster when repeatedly thawed. Short-term use at 4°C for one to two weeks is acceptable when handled aseptically. The unconjugated format leaves the choice of secondary to the user: HRP conjugates for chemiluminescent Western blot and chromogenic immunohistochemistry, alkaline phosphatase conjugates for chromogen-stable workup, or fluorophore conjugates for immunofluorescence colocalization studies. Investigators studying cytoskeletal remodeling, dendritic spine plasticity in neurons, neutrophil chemotaxis, host-pathogen interactions involving Listeria or Shigella actin comet tails, and invadopodium dynamics in metastatic cell models will find this reagent useful.
