From Antibodies.com, this unconjugated monoclonal primary antibody targets ATP citrate lyase (ACLY), the cytosolic homotetrameric enzyme that converts citrate plus CoA into acetyl-CoA and oxaloacetate using ATP hydrolysis. The reagent is supplied as a 200 µl aliquot of mouse IgG in PBS buffer (pH 7.3) containing 0.02% sodium azide and 50% glycerol as a cryoprotectant.
For western blot, the antibody has been validated at working dilutions between 1:500 and 1:2000, detecting ACLY at an observed molecular weight of 121 kDa from human, mouse, or monkey cell and tissue lysates. Validation data used 25 µg protein per lane with 3% nonfat dry milk in TBST as blocking buffer and an HRP-conjugated goat anti-mouse IgG secondary. For immunocytochemistry, the recommended dilution range is 1:50 to 1:200, and for flow cytometry the suggested dilution is 1:20 to 1:50, supporting quantitative single-cell measurement of endogenous ACLY in human, mouse, and monkey cell populations.
Because the antibody is unconjugated, laboratories pair it with HRP-, fluorophore-, or biotin-coupled secondary antibodies compatible with mouse IgG, retaining flexibility across detection platforms. The unconjugated format also supports signal amplification strategies and custom labelling protocols. Researchers use the reagent for orthogonal confirmation of ACLY status at the protein level after siRNA, shRNA, or CRISPR interventions, and to monitor changes in ACLY expression under metabolic or pharmacological challenge, including Warburg-effect-driven lipogenesis in cancer cells and SREBP- or ChREBP-mediated transcriptional control of lipid biosynthesis.
Storage: store at -20 °C and avoid repeated freeze-thaw cycles; the 50% glycerol buffer component helps preserve antibody activity in the vial. The product is for research use only.
