Allprep RNA Protein Kit (50)

This spin-column kit from QIAGEN enables the simultaneous purification of total RNA and native protein from a single sample of cultured cells. It uses silica-based technology with manual centrifugation processing, requiring approximately 40 minutes per preparation from a sample amount of 700 µl. The elution volume is 30–50 µl.

The kit is designed for downstream applications including PCR, real-time PCR, blotting, and microarray analysis. It is particularly useful for validating siRNA-mediated knockdown at both mRNA and protein levels, as obtaining RNA and protein from the same sample improves the reliability of knockdown analysis while reducing the number of cell cultures required. The kit forms part of QIAGEN's range of closely integrated products for RNAi and gene expression analysis.

In validation studies, total RNA purified from HeLa cells showed an rRNA ratio (28S/18S) of 2.04. Total protein analyzed by western blot using antibodies against Lamin (nuclear protein) and TIM23 (mitochondrial protein) demonstrated that the protein fraction contains proteins from subcellular compartments. The kit has been optimized for cultured cells, with a maximum of 6 × 10⁵ cells per preparation; using larger cell numbers may reduce RNA as well as protein yield and quality.
