QIAGEN offers a system for the simultaneous purification of genomic DNA and total RNA from a single animal cell or tissue sample. The AllPrep DNA/RNA Mini Kit processes a single lysate sequentially through an AllPrep DNA spin column to isolate DNA, then through an RNeasy spin column to isolate RNA, capturing both nucleic acid types without splitting the sample.

The procedure uses a guanidine-isothiocyanate-containing lysis buffer that immediately inactivates DNases and RNases, helping to ensure isolation of intact nucleic acids. Genomic DNA has an average length of 15–30 kb depending on homogenization conditions, and RNA molecules longer than approximately 200 nucleotides are enriched. The kit is compatible with small amounts of a wide range of animal cells and tissues and supports parallel processing of multiple samples in less than 40 minutes, eliminating the need for phenol/chloroform extraction or CsCl step-gradient ultracentrifugation.

Purified DNA is suitable for next-generation sequencing, PCR, Southern, dot and slot blot analyses, comparative genome hybridization, and genotyping or SNP analysis. Purified RNA is suitable for RNA-seq, RT-PCR, quantitative real-time RT-PCR, cDNA synthesis, Northern, dot and slot blot analyses, primer extension, poly A+ RNA selection, RNase/S1 nuclease protection, differential display, and microarray applications. The kit is intended for research use only and should be stored dry at room temperature (15–25 °C), where it is stable for at least 9 months under these conditions.
