Description

From QIAGEN's primary sample technology portfolio, the AllPrep Bacterial/Fungal DNA/RNA/Protein Kit is designed for extraction of nucleic acids from bacterial and fungal cell cultures. The kit isolates total nucleic acids and cellular proteins from cultured Gram-negative and Gram-positive bacteria as well as cultured fungi, using bead-beating combined with silica spin-filter technology. Two bead options are supplied depending on the sample: 0.1 mm glass beads for bacterial lysis and 0.5 mm glass beads for fungal lysis, with processing carried out by chemical lysis and/or bead beating.

Cells are lysed in glass bead tubes, then nucleic acids and proteins are solubilized and mixed with a binding solution. DNA binds to the first spin column; the flow-through is combined with a further solution to capture total RNA on a second spin column, while the last flow-through retains protein. Each analyte is washed on its respective column and eluted separately. The recommended sample input is 1.8 ml of microbial culture, with QIAGEN recommending no more than 1×10⁹ bacterial cells per sample, or 1.8 ml of fungal culture. Throughput is 1–24 samples per run, with a typical prep time of around 36 minutes.

Eluted DNA and RNA are suitable for downstream applications such as qPCR, RT-PCR and next-generation sequencing, while eluted protein is ready for 1D SDS-PAGE and mass spectrometry following in-gel trypsin digestion. Some downstream uses, including 2D SDS-PAGE and solution-phase proteolytic digestion for mass spectrometry, may require an additional detergent-removal step after protein elution. For storage, the spin filters should be kept at 4°C, while the remaining kit reagents and components can be stored at room temperature (15–30°C).
