Description

Circulating tumor cells (CTCs) from whole blood can be enriched for downstream molecular characterization in colon cancer research using QIAGEN's AdnaTest ColonCancerSelect. The system employs an immunomagnetic cell-selection approach in which antibodies directed against epithelial and tumor-associated antigens are conjugated to magnetic beads, enabling labeling and capture of tumor cells. The product is intended for molecular biology applications and is used together with AdnaTest ColonCancerDetect to analyze colon-cancer-associated gene expression in the enriched cells; it is not intended for the diagnosis, prevention, or treatment of a disease.

Labeled cells are extracted using a magnetic particle concentrator (AdnaMag-L or AdnaMag-S) and are subsequently lysed. The workflow follows a two-step "select and detect" process that generates results within 5 hours. The cell lysate produced is then used for downstream analysis with AdnaTest ColonCancerDetect, including mRNA isolation, reverse transcription, and multiplex PCR characterization of tumor-associated markers.

In spiking experiments, 5 tumor cells in 5 ml of whole blood are detected at a recovery rate of at least 90%, and the specificity of detection is reported as at least 90%. Kit components are stored at 2–8°C and must not be used beyond the expiration date. Each lot is tested against predetermined specifications under QIAGEN's ISO-certified Quality Management System.
