Description

This cell-permeable small molecule from STEMCELL Technologies is a direct, allosteric activator of AMP-activated protein kinase (AMPK). It produces a 4.1-fold stimulation of AMPK with an EC₅₀ of 116 nM, potentially by inhibiting dephosphorylation on Thr172, and specifically activates β1 subunit–containing AMPK heterotrimers independently of upstream kinases. The compound also inhibits Na⁺/K⁺-ATPase.

A769662 (CAS 844499-71-4; molecular formula C₂₀H₁₂N₂O₃S; molecular weight 360.4 g/mol) is supplied as a crystalline solid with a stated purity of ≥ 98%. Reported solubility includes ~20 mg/mL in DMSO and ~0.5 mg/mL in a 1:1 solution of DMSO:PBS (pH 7.2) or in ethanol and DMF. Stock solutions in DMSO can be prepared at concentrations up to 55 mM, while absolute ethanol supports up to 1 mM; the compound has low solubility in aqueous media.

Storage recommendations from the supplier include keeping the product at −20 °C, protected from prolonged exposure to light, where it is stable for 12 months from the date of receipt. Published research applications include stem cell maintenance and self-renewal, reprogramming, cancer research, and metabolism studies. The product is classified as a laboratory chemical intended for research use only.
