Description

This small molecule from STEMCELL Technologies is a membrane-permeable cAMP derivative that activates cAMP-dependent protein kinase. It is described by the manufacturer as a cAMP pathway activator and is noted for long-acting effects owing to its resistance to cAMP phosphodiesterase (Schwede et al.). The compound has been used to study calcium-mediated pathways, with an IC₅₀ of 0.84 mM reported in macrophage studies (Xaus et al.).

Provided as a crystalline solid, the product has a stated purity of ≥ 95%. Its molecular formula is C₁₀H₁₁BrN₅O₆P with a molecular weight of 408.1 g/mol, and the CAS Number is 23583-48-4. Alternative names listed for the compound include 8-BrcAMP, 8-Bromoadenosine 3',5'-cyclic monophosphate, and NSC 171719. Reported solubility in PBS (pH 7.2) is ≤ 7.4 mM. The manufacturer recommends storage at -20°C, protected from prolonged light exposure, and notes stability for 12 months from date of receipt when stored with a desiccant.

Published applications for 8-Bromo-cAMP span several research areas. In reprogramming, it has been shown to improve the reprogramming efficiency of human neonatal foreskin fibroblast (HFF1) cells when combined with Valproic Acid (Wang & Adjaye), and the manufacturer lists reprogramming as a primary application. In immunology, the compound inhibits M-CSF-dependent proliferation of macrophages (Xaus et al.) and protects neutrophils against TNF-α-induced apoptosis (Krakstad). In cancer research, it induces a proliferative response in an IL-3-dependent leukemic cell line (Barge et al.) and induces membrane depolarization in pancreatic cancer cell lines (Sorio et al.).
