6x DNA Loading Dye Buffer Double Blue

This loading dye buffer from Solis BioDyne is intended for loading DNA samples, including PCR products and restriction fragments, onto agarose or polyacrylamide gels.

The reagent contains bromophenol blue, Xylene Cyanole FF, TRIS-HCl, EDTA and Ficoll. In a 1% agarose gel, bromophenol blue comigrates with approximately 300 bp fragments in 1× TBE or ~400 bp fragments in 1× TAE, while Xylene Cyanole FF comigrates with approximately 3500 bp fragments in 1× TBE or ~5000 bp fragments in 1× TAE. No interaction with the borate on TBE gels is observed, and the Ficoll component alters local pH.

The recommended protocol is to mix 1 volume of the Double Blue Dye with 5 volumes of the DNA sample before loading. Shipping at room temperature has no detrimental effects on the quality of the reagent. It can be stored at room temperature or at 4ºC, and for longer periods at -18ºC to -28ºC.
