6x DNA Loading Dye Buffer Blue

From Solis BioDyne, this 6× concentrate blue loading dye buffer is designed to prepare DNA samples before electrophoresis. The reagent is a slightly viscous blue liquid composed of bromophenol blue, TRIS-HCl, EDTA and Ficoll, and is intended for research use only / in vitro use.

It is used to load DNA samples such as PCR products and restriction fragments onto agarose or polyacrylamide gels. In a 1% agarose gel the bromophenol blue dye comigrates with approximately 300 bp fragments in 1× TBE buffer or approximately 400 bp fragments in 1× TAE buffer. The formulation is reported not to interact with borate on TBE gels, while the Ficoll component alters the local pH. The mixture has a pH of 7.0–9.0. Typical use involves mixing one volume of the dye with five volumes of the DNA sample before loading onto the gel.

The buffer is supplied as a non-hazardous mixture and is not classified under CLP for physical, health or environmental hazards. Routine storage is at room temperature or 4 °C, with longer-term storage at −20 °C; the SDS also indicates storage between −28 °C and −18 °C. Users are advised to handle the product with appropriate laboratory protective equipment and to wash affected skin or eyes with water on contact.
