Description

QIAGEN's lipid-based transfection reagent is formulated for efficient delivery of RNA into eukaryotic cells. The ready-to-use formulation combines a lipid component with a specific RNA-condensing enhancer and an optimized buffer, so that RNA molecules are first condensed by the enhancer and then coated with the reagent for effective transfer into cells. All components are supplied as ready-to-use solutions, and no controls are included.

Quality control testing covers the absence of RNases, batch-to-batch consistency, and low endotoxin levels (≤10 EU/ml), removing variables that could otherwise compromise RNA transfection efficiency. The reagent supports transfection with RNA for direct studies of RNA function and is described by QIAGEN as enabling efficient transfection of neuronal cells. It is suitable for transient transfection as well as co-transfection applications.

Cell lines successfully transfected include 293T, Jurkat and Vero cells, and the reagent can also be used to transfect RNA into human primary fibroblasts, rat cardiomyocytes and rat neural cells. To form transfection complexes, RNA is mixed with Enhancer R and Buffer EC-R and incubated at room temperature for 5 minutes, after which the reagent is added and the mixture is incubated for a further 5–10 minutes. The complexes are then mixed with medium and added directly to the cells, followed by a 3-hour incubation and a medium change. High-purity RNA is recommended for optimal results, and QIAGEN highlights RNA purified with RNeasy. Because RNA is transfected rather than DNA, expression occurs without transcription and is independent of a promoter, with protein expression typically appearing sooner than after DNA transfection.
