Description

Solis BioDyne offers a heat-labile Uracil-N-Glycosylase enzyme designed to eliminate carryover contamination in PCR and LAMP workflows. The enzyme removes uracil from single- or double-stranded DNA by catalysing the hydrolysis of the N-glycosylic bond and leaving an abasic site, so that degraded dU-DNA cannot serve as a template for further amplification.

The protein sequence originates from the bacterial genus Salinivibrio, which is frequently found in hypersaline environments. Through Solis BioDyne's proprietary and patented Stability TAG technology, the enzyme is exceptionally stable at room temperature, supporting reaction set-up and shipment without ice. It is inactivated by a 5-minute heat treatment at 70°C, and no reactivation is detected after the heat-treated enzyme is stored at 4°C or 25°C for 48 hours.

With a fast reaction time and tolerance to common inhibitors, the enzyme is also compatible with Sanger sequencing. Beyond carryover prevention, it serves as an enhancer of cloning efficiency for PCR products and supports site-directed mutagenesis, protein-DNA interaction studies, glycosylase-mediated single nucleotide polymorphism detection (GMPD), DNA repair and mutation studies, and SNP genotyping. A glycerol-free formulation is available upon request.
