QIAGEN's 3' targeted RNA sequencing chemistry delivers single-tube library construction built around a 12-base unique molecular identifier (UMI) and integrated Cell ID tagging, enabling accurate transcript counting and PCR duplicate removal without separate index transfer steps. The pre-dried, single-use 96-index plate C ships as part of Set C indexing within the broader 384-sample workflow, with breakable plate geometry that allows processing of partial plates in batches of 96.

The kit accepts 1 to 100 cells per reverse transcription reaction via direct cell lysis, with a minimum of 24 cells recommended to be multiplexed per sample index when working with single cells. For purified RNA inputs, the supported range spans 10 pg to 1 ng of total RNA. A 20 µl aliquot is transferred to a real-time PCR plate for the quantitative universal PCR step, allowing cycle-by-cycle monitoring of library amplification before pooling. The LNA-enhanced anchored oligo-dT RT primer carries the integrated Cell ID and UMI, so all 384 RT reactions can be consolidated into a single 2 ml microcentrifuge tube after reverse transcription.

Compatible libraries are produced for Illumina sequencing platforms, with 4 nM denatured libraries recommended as sequencing input and 12 pM loading concentration for MiSeq V3 runs. Kit components include EZ Reverse Transcriptase, HotStarTaq DNA Polymerase, QIAseq Beads, SPE Buffer, and indexed primers pre-aliquoted in single-use plates. Store at -20 °C, protect from light, and avoid repeated freeze-thaw cycles to preserve enzyme activity. Intended for research use only.
