PyroMark Q24 Validation Oligo

QIAGEN's PyroMark Q24 Validation Oligo is designed for performance confirmation of the PyroMark Q24 system. The product consists of two biotinylated oligonucleotides that differ in sequence at one position, synthesized as either A or G. By mixing the two oligonucleotides in different proportions, a variable position is created: C or T is incorporated upon sequencing, and the variable position is analyzed as %C. Replicates of these mixes are used to determine linearity, bias, and repeatability, constituting the performance test of the system. The limits of the proportions of the two mixes, 5% and 95%, have been selected to coincide with generally-accepted limits for reliable quantification as determined by in-house evaluation and published data.

Each oligonucleotide can form an internal stem-loop structure that enables self-priming for extension by DNA polymerase, eliminating the need for a sequencing primer in the Pyrosequencing reaction. The kit includes PyroMark Q24 Validation Oligo 5% (20 μM, 70 μl), PyroMark Q24 Validation Oligo 95% (20 μM, 70 μl), and Dilution Buffer (2 x 1.7 ml). The product should be stored at –20°C and remains stable until the expiration date under these conditions; repeated thawing and freezing (>4 times) should be avoided.

The PyroMark Q24 Validation Oligo is intended for molecular biology applications. It is neither intended for the diagnosis, prevention, or treatment of a disease, nor has it been validated for such use either alone or in combination with other products. Each lot is tested against predetermined specifications in accordance with QIAGEN's ISO-certified Quality Management System to ensure consistent product quality.
