Description

This single-use vessel from STEMCELL Technologies is part of the PBS-MINI compact bioreactor system, designed for high-throughput, 3D suspension culture of human pluripotent stem cells (hPSCs) and other cell types. Vessels are available with and without a bottom port.

The 0.1 MAG vessel supports hPSC expansion at culture volumes of 80 – 100 mL, with a recommended agitation rate of 35 – 40 rpm, and is used with the PBS-MINI Bioreactor Base Unit. According to manufacturer data, hES (H9) and hiPS (WLS-1C) cell lines seeded at 8 x 10⁴ viable cells/mL in 100 mL and cultured for 4 days using mTeSR™3D showed consistently high daily fold expansion and end-of-passage viability, comparable to Nalgene™ filter bottle controls. hPSC aggregates expanded in the vessel exhibit typical suspension morphology — mostly spherical with clear edges and a dimpled appearance — typically 350 – 400 µm in size. In multi-vessel comparisons, the 0.1 MAG vessel was among those recommended for robust suspension culture scale-up, with a consistent mean daily fold expansion above 1.2.

The vessel also supports 3D hematopoietic differentiation workflows. In an evaluation of the STEMdiff™ Hematopoietic Kit, hPSCs differentiated in PBS-MINI 0.1 L vessels produced HPCs that expressed the pan-hematopoietic marker CD43 at levels exceeding 90%, with an average yield of nearly one million CD34⁺CD45⁺ cells per mL of culture volume — equivalent to a total output of 100 million CD34⁺CD45⁺ HPCs in one 0.1 L experiment. These HPCs were shown to be functional in colony-forming unit (CFU) assays, producing comparable CFU-GEMM, BFU-E, and CFU-GM colonies to those generated in standard 2D and other 3D platforms. All cultures should be maintained in an incubator at 37 °C with 5% CO₂.
