QIAGEN's premium-grade LNA-optimized oligonucleotide is intended for miRNA functional studies, where it simulates naturally occurring mature miRNAs to enable gain-of-function investigations. The design features a distinctive triple-strand RNA configuration: an unmodified guide strand whose sequence corresponds exactly to the miRBase annotation, paired with two shorter LNA-enhanced passenger strands. This segmented passenger architecture ensures that only the intended miRNA strand is incorporated into the RNA-induced silencing complex (RISC), so off-target effects from the passenger strands are minimal and phenotypic changes can be ascribed specifically to the targeted miRNA.

Incorporation of locked nucleic acid (LNA) chemistry raises the duplex melting temperature by 2–8°C for every LNA monomer added, conferring high affinity and the ability to design short oligonucleotides with strong target binding. The result is improved sensitivity and specificity, including robust performance across miRNA targets of widely varying GC content and specific discrimination of closely related miRNA family members.

The product is intended for molecular biology applications and is not intended for the diagnosis, prevention, or treatment of disease. Each lot is tested against predetermined specifications under QIAGEN's ISO-certified Quality Management System. Vials are shipped at room temperature and should be stored at –15 to –30°C or below, remaining stable for at least 6 months after the shipping date. The premium grade offers enhanced purification and LNA modifications for demanding applications such as high-sensitivity functional assays, mechanistic validation, and precise dose-response studies.
