miRCURY LNA miRNA Mimic (20)

A line of LNA-optimized oligonucleotides from QIAGEN supports miRNA functional studies. Supplied as miRCURY LNA miRNA Mimics, the products are intended for molecular biology applications and are not intended for the diagnosis, prevention or treatment of a disease.

The mimic design uses three RNA strands: an unmodified miRNA guide strand whose sequence matches the miRBase annotation, and two shorter LNA-modified passenger strands complementary to the guide strand. Because the passenger strand is split, only the miRNA strand is loaded into the RNA-induced silencing complex (RISC), while the two passenger strands are too short to act as miRNAs and are rapidly degraded after displacement. The sequence, length and LNA spiking pattern of the segmented passenger strands have been optimized using an empirically derived design algorithm, so phenotypical changes can be safely ascribed to the miRNA simulated by the mimic. The mimics simulate the natural functions of endogenous miRNAs and are primarily used in gain-of-function studies, enabling assessment of biological consequences such as cell proliferation, differentiation or apoptosis, as well as validation of miRNA targets in combination with inhibitors and target site blockers.

Negative controls are available with guide strands that have no homology to any known miRNA or mRNA sequence in mouse, rat or human, and that have been tested for adverse effects in multiple cell lines. Each lot is tested under QIAGEN's ISO-certified Quality Management System to ensure consistent product quality. Mimics are shipped at room temperature; unopened vials should be stored at –15 to –30ºC or below, fluorescently labeled mimics should be protected from light to avoid bleaching, and products stored in this manner remain stable at least 6 months after the shipping date.
