Locked 5-methylcytosine residues are introduced into synthetic oligonucleotides using the LNA-mC(Bz) Amidite, a phosphoramidite building block supplied by QIAGEN for solid-phase synthesis on standard DNA/RNA synthesizers. The bicyclic LNA sugar locks the ribose in the C3'-endo conformation, which raises duplex melting temperature by several degrees Celsius per modification and improves single-base mismatch discrimination during probe hybridization.

Each 250 mg vial contains the N4-benzoyl-protected, DMTr-off phosphoramidite in a sealed format ready for coupling on controlled-pore glass or polystyrene supports, with typical coupling efficiency above 97% under standard activator conditions. The 5-methylcytosine base reproduces the epigenetic methylation pattern seen at CpG dinucleotides in vertebrate DNA, making the reagent especially useful for methylation-specific PCR primer design, bisulfite-conversion sequencing workflows, miRNA in situ hybridization panels, and antisense oligonucleotide constructs where nuclease resistance matters.

Storage at -20 °C in a desiccated environment, protected from light and moisture, preserves coupling performance; allow the vial to equilibrate to room temperature before opening to prevent condensation on the dry solid.
