Description

A dye-based qPCR mix from Solis BioDyne (based in Tartu, Estonia) designed for real-time quantitative PCR. The mix contains all components needed for the reaction except sample (DNA/RNA template) and primers, and is described as delivering accurate and highly sensitive results. It incorporates a specially developed SolisGreen dye that is stated to be environmentally friendly and safe to use.

A passive reference based on ROX dye is included, making the product compatible with both ROX-dependent and ROX-independent qPCR cyclers. The mix uses HOT FIREPol DNA Polymerase, which is activated by a 10 min incubation step at 95°C, helping to prevent the extension of non-specifically annealed primers and primer-dimers formed at low temperatures during reaction setup. The formulation is reported to be exceptionally stable at higher temperatures, enabling ice-free shipping.

Manufacturer data shows that stability testing delivered high fluorescence levels and reproducibility after 2 weeks at 37°C and after 1 month at room temperature (25°C), compared with a reference kept at -20°C, using a TUBA8 target across four 10-fold dilutions (20 pg to 20 ng) of human gDNA on a QuantStudio 6 Pro qPCR cycler. A separate comparison indicates approximately two times brighter fluorescence signal than HOT FIREPol EvaGreen qPCR Supermix when amplifying a B2M target across five 10-fold cDNA dilutions (0.01 ng to 0.1 µg). Documented applications include gene expression analysis, absolute quantification, pathogen detection and quantification, low-copy gene detection, and cell-free DNA analysis. According to the manufacturer, the mix is also compatible with most qPCR platforms and offers improved detection of low-copy targets.
