Description

From QIAGEN, this lyophilized, preannealed siRNA reagent is intended for efficient RNAi analysis one gene at a time. Design uses the proprietary HP OnGuard siRNA Design, which combines BioPredsi neural-network technology with homology analysis against an up-to-date nonredundant sequence database, asymmetry to favour antisense-strand entry into RISC, 3' UTR/seed-region analysis to limit off-target effects, single-nucleotide-polymorphism avoidance through the RefSNP database, and screening for interferon-inducing sequence motifs. Genome-wide Affymetrix GeneChip analysis and current NCBI databases inform target sequence selection.

The reagent is supplied lyophilized in tubes and is ready for use after resuspension in the sterile RNase-free water provided. It is available for human, mouse, and rat genes, and full sequence information is provided at no extra charge to support analysis and publication. Modification options include fluorescent labels (Alexa Fluor, fluorescein, rhodamine, Cy3, and Cy5) and chemical modifications such as amino linkers, thio linkers, and phosphate modifications. For transfection, QIAGEN recommends HiPerFect Transfection Reagent for low-throughput work and HiPerFect HTS Reagent for high-throughput applications. Items ship lyophilized at room temperature.

Designed for transfection, validation, and functional assays, the reagent is compatible with common transfection reagents and laboratory automation. Handling recommendations include storage at -20°C and avoiding repeated freeze-thaw cycles. siRNAs produced by the HiPerformance algorithm typically achieve greater than 70% knockdown, and thousands of human siRNAs have been experimentally verified by real-time RT-PCR analysis to deliver at least 70% knockdown. A one-time-only replacement offer applies: if at least two siRNAs ordered for the same target gene fail to provide at least 70% knockdown, QIAGEN will supply two additional siRNAs free of charge on submission of supporting data demonstrating the siRNA failed to knock down the target gene by at least 70% at the mRNA level under appropriate transfection conditions, including transfection efficiency data, quantitative silencing data, and data showing at least 70% knockdown of a positive control, within 6 months of delivery. QIAGEN recommends testing at least two distinct siRNAs per gene to confirm specificity.
